Just make sure they are using the same driver (i.e.:
ivtv (used by the cards that do hardware encoding the
Hauppage PVR cards, and Adaptec AVC-2410) or the v4l
(used by the cheap/nasty tuner cards)).

With recording you don't directly tell MythTV which
tuner to use. You can specify preferences. Say tuner 0
is tied to an antenna that gets fantastic reception,
but only a handfull of channels, tuner 1 is tied to
cable TV, so-so quality but a huge number of channels.
You can specify that tuner 0 gets a +1 priority so
given a choice MythTV should record from tuner 0, but
if something is only available via tuner 1, it will go
that way.

My own set-up, both cards are tied to the same cable
TV signal and both cards are at priority 0 (the
default).
